RESUMO

Several psychodid flies are commonly associated with human-inhabited environments and have been increasingly implicated in cases of human myiasis. However, the basic biology of psychodid larvae is not well-suited for survival in the human intestinal or urogenital tract, making true, prolonged myiasis unlikely. In this review, we performed a systematic literature review of published cases of purported myiasis caused by psychodid flies, their identification, associated clinical findings, and treatment. We also discuss the anatomy and lifecycle of psychodid flies in relation to their purported ability to use human tissue as a nutritive source and survive in the human alimentary or urogenital tracts. Based on the range of non-specific and varied reported clinical manifestations, lack of observed collections, life cycle patterns of psychodid flies, the mechanics of their mouthparts, and breathing requirements, we conclude that most cases likely represent incidental findings, or in rare cases possibly pseudomyiasis, rather than true myiasis, and provide recommendations for clinical evaluation and reporting so that disease misclassification and unnecessary therapy do not occur.

The recent geographic spread of Leishmania infantum along the borders of Argentina, Brazil and Paraguay has been highlighted. In our previous study, Lutzomyia longipalpis was found in 55 of 123 patches surveyed, and in some patches, sandflies were found at higher densities, forming hotspots. Based on the One Health approach, we investigated the seasonality of the vector, the presence of parasite DNA, and the environmental factors that contribute to vector and parasite dispersal in these previously described hotspots in Foz do Iguaçu, Brazil. Entomological surveys were conducted monthly for one year. Fourteen hotspots peridomicile and six intradomicile were sampled. PCR was used to assess the prevalence of Leishmania DNA in sandflies. Zero-inflated negative binomial regression was used to determine the association of micro- and mesoscale environmental variables with the occurrence and abundance of the three most abundant sandfly species sampled. A total of 3543 species were captured, with Lutzomyia longipalpis being the predominant species (71.78%) of the 13 species found. Evandromyia edwardsi, Expapillata firmatoi, Micropygomyia ferreirana and Pintomyia christenseni were reported for the first time in the region. NDVI, distance to water, precipitation, west-to-east wind, wind speed, maximum and minimum relative humidity, and sex were significant variables associated with vector presence/abundance in the environment. Vector presence/abundance in the peridomicile was associated with precipitation, altitude, maximum temperature, minimum and maximum relative humidity, west-to-east wind, wind speed, and sex. Leishmania DNA was detected in an average of 21% of Lu. longipalpis throughout the year. Vector abundance is concentrated in urban and peri-urban areas, with some specimens present in different parts of the city and some sites with high vector abundance. This distribution suggests that the risk of actual contact between humans and parasite vectors in urban areas during the epidemic period is associated with patches of peri-urban vegetation and then extends into urban areas.

Background: Leishmaniases are a group of vector-borne zoonotic diseases of public health relevance within the tropical and subtropical regions of the world. The state of Yucatan is a vulnerable and receptive area to localized cutaneous leishmaniasis (LCL) due to its proximity to the high-transmission endemic states of Campeche and Quintana Roo. Autochthonous cases of LCL caused by Leishmania (Leishmania) mexicana have been documented in the state, showing a geographical expansion of the disease. Materials and Methods: Using CO2-supplemented Centers for Disease Control and Prevention light traps and Shannon traps, we captured anthropophilic sandflies in the surroundings of a locality with recent records of autochthonous cases of LCL. Sandflies carrying Leishmania DNA were evidenced using PCR. Results: A total of 140 Phlebotominae (Diptera: Psychodidae) females of four species were captured: Lutzomyia (Tricholateralis) cruciata (Coquillett), Psathyromyia (Psathyromyia) shannoni (Dyar), Lutzomyia (Lutzomyia) longipalpis (Lutz and Neiva), and Dampfomyia (Coromyia) deleoni (Fairchild and Hertig). Molecular results showed that 6.1% (95% confidence interval [CI] = 2.3-12.9%) of Lu. cruciata and 43.8% (95% CI = 19.8-70.1%) of Pa. shannoni showed evidence of carrying L. (L.) mexicana DNA. Conclusion: We provide evidence of anthropophilic sandflies carrying L. mexicana DNA in a municipality with recorded autochthonous cases of LCL caused by this parasite species in the state of Yucatan, suggesting the emergence of new focus of LCL in Mexico.

Geographical variation in abundance and composition of pollinator assemblages may result in variable selection pressures among plant populations and drive plant diversification. However, there is limited knowledge on whether differences in local visitor and pollinator assemblages are the result of site-specific strategies of plants to interact with their pollinators and/or merely reflect the pollinator availability at a given locality. To address this question, we compared locally available insect communities obtained by light-trapping with assemblages of floral visitors in populations of Arum maculatum (Araceae) from north vs. south of the Alps. We further investigated whether and how the abundance of different visitors affects plants' female reproductive success and examined the pollen loads of abundant visitors. Local insect availability explained inter-regional differences in total visitor abundance, but only partly the composition of visitor assemblages. Northern populations predominantly attracted females of Psychoda phalaenoides (Psychodidae, Diptera), reflecting the high availability of this moth fly in this region. More generalized visitor assemblages, including other psychodid and non-psychodid groups, were observed in the south, where the availability of P. phalaenoides/Psychodidae was limited. Fruit set was higher in the north than in the south but correlated positively in both regions with the abundance of total visitors and psychodids; in the north, however, this relationship disappeared when visitor abundances were too high. High pollen loads were recorded on both psychodids and other Diptera. We demonstrate for the first time that the quantitative assessment of floral visitor assemblages in relation to locally available insect communities is helpful to understand patterns of geographical variation in plant-pollinator interactions. This combined approach revealed that geographical differences in floral visitors of A. maculatum are only partly shaped by the local insect availability. Potential other factors that may contribute to the geographical pattern of visitor assemblages include the region-specific attractiveness of this plant species to flower visitors and the population-specific behavior of pollinators.

Background: The Psychodinae of the Afrotropical Region remain poorly understood. Slightly under 200 species have been described, but many countries have received very little attention from collectors and even countries where significant collection efforts have taken place have rarely had their collections studied in detail by specialist taxonomists. New information: Perithreticusneglectus sp. n. is described from the West Usambara Mountains, Tanzania, based on a male specimen collected in 1990. The new species is similar to Perithreticusanderseni Kvifte, 2015, which occurs in the same forest reserve, but can be separated by several genitalic characters, including the hypandrium well-developed with sclerotised anterior and posterior margins, gonocoxites narrower, the gonostyles with the slender apex shorter, the parameres shorter without pronounced basolateral projections and the surstylus with slightly fewer tenacula. The world fauna of Perithreticus now comprises five described species, of which two occur in the Afrotropical Region.

Floral scent is vital for pollinator attraction and varies among and within plant species. However, little is known about how inter-individual variation in floral scent affects the abundance and composition of floral visitor assemblages within populations. Moreover, for deceptive plants it is predicted that intra-population variation in scent can be maintained by negative frequency-dependent selection, but empirical evidence is still lacking. To investigate the ecological and evolutionary relations between inter-individual scent variation (i.e., total emission and composition) and floral visitors in deceptive plants, we studied floral scent, visitor assemblages, and fruit set in two populations of fly-pollinated (Psychodidae, Sphaeroceridae; Diptera) and deceptive Arum maculatum from Austria (JOS) and northern Italy (DAO). By correlating individual data on floral scent and visitor assemblages, we show that inter-individual variation in floral scent partly explains variation in visitor assemblages. The quantity of floral scent emitted per individual correlated positively with visitor abundance in both populations but explained visitor composition only in DAO, where strongly scented inflorescences attracted more sphaerocerid flies. However, in each population, the composition of floral scent did not correlate with the composition of floral visitors. There was also no evidence of negative frequency-dependent selection on floral scent. Instead, in JOS, more frequent scent phenotypes attracted more pollinators and were more likely to set an infructescence than rarer ones. Our results show that floral scent, despite being key in pollinator attraction in A. maculatum, only partly explains variation in pollinator abundance and composition. Overall, this study is the first to shed light on the importance of inter-individual variation in floral scent in explaining floral visitor assemblages at the population level in a deceptive plant species.

Leishmania donovani is the causative agent of historically anthroponotic visceral leishmaniasis (VL) on the Indian subcontinent (ISC). L. donovani is transmitted by the sand fly species Phlebotomus argentipes. Our collaborative group and others have shown that sand flies trapped outside in endemic villages have fed on cattle and dogs in addition to people. Domestic animals are reservoirs for L. donovani complex spp., particularly L. infantum, in other endemic areas. Multiple studies using quantitative PCR or serological detection methods have demonstrated that goats, cattle, rats and dogs were diagnostically positive for L. donovani infection or exposure in eastern Africa, Bangladesh, Nepal and India. There is a limited understanding of the extent to which L. donovani infection of domestic animals drives transmission to other animals or humans on the ISC. Evidence from other vector-borne disease elimination strategies indicated that emerging infections in domestic species hindered eradication. The predominant lesson learned from these other situations is that non-human reservoirs must be identified, controlled and/or prevented. Massive efforts are underway for VL elimination on the Indian subcontinent. Despite these herculean efforts, residual VL incidence persists. The spectre of an animal reservoir complicating elimination efforts haunts the final push towards full VL control. Better understanding of L. donovani transmission on the Indian subcontinent and rigorous consideration of how non-human reservoirs alter VL ecology are critical to sustain elimination goals.

ABSTRACT The municipality of Paracambi (Rio de Janeiro, Brazil) reports sporadic cases of American cutaneous leishmaniasis (ACL). Previous studies detected Nyssomyia intermedia (Diptera: Psychodidae) as the main vector in the region, but its spatial distribution and the presence of other vector species have not been evaluated. This study aims at filling this knowledge gap, by studying the ecology of sand flies, their spatiotemporal distribution, and correlation with land use/cover. Two campaigns of monthly sand fly collections using light traps and manual captures were conducted in 1992-1994 and 2001-2003. Females were dissected to detect natural Leishmania infections. The spatial distribution of sand flies was assessed using kernel density maps. Correlations with land use/cover were evaluated by extracting satellite imagery data around the capture points. A total of 17,232 sand flies from 13 species were captured. Medically important species included Ny. intermedia, Migonemyia migonei, Pintomyia fischeri and Ny. whitmani. No Leishmania-infected females were detected. Highest densities were detected in the peri-urban areas Cascata and Sabugo, and in rural areas São José and Mutirão. Ny. intermedia had statistically significant correlations with pasture and agricultural areas. Present results strengthened that Ny. intermedia and Mg. migonei are the main local ACL vectors. Correlations with land use evidence the association between ACL and anthropic environmental change.

ABSTRACT OBJECTIVE To perform an entomological survey, evaluating the circulation of Leishmania spp. in sand flies captured from the new foci of visceral leishmaniasis (VL) in the coastal region of São Paulo state. METHODS Sand flies were captured from November 2016 to September 2018 using light traps of the Centers for Disease Control (CDC), in the neighborhood where VL cases were reported. Leishmania spp. circulation was evaluated by gut dissection and molecular analysis of the females captured. RESULTS Nyssomyia intermedia was the more frequent species (90,7%) within the 1,203 sand flies captured. We found no flagellates in dissected females, but two pools containing females of Ny. intermedia presented DNA of L. infantum. CONCLUSION Our results suggest that Ny. intermedia might be involved in the establishment of this new VL focus in Sao Paulo. However, before incriminating this species as a L. infantum vector, further studies should investigate other vectorial capacity parameters, including competence, survival, and feeding habits.

ABSTRACT Background: The American cutaneous leishmaniasis (ACL) is expanding in peri-urban environments. Methods: An entomological survey was conducted in the area of the occurrence of an autochthonous urban case of ACL. Sandflies and a parasitological slide of the human case were submitted for molecular diagnosis. Results: Nyssomyia whitmani and Ny. antunesi were the most frequently collected species. Ny. whitmani and Bichromomyia flaviscutellata were positive for Leishmania guyanensis and L. lainsoni, respectively. The human case tested positive for L. lainsoni. Conclusions: Sandflies and Leishmania parasites present in urban forest may occur frequently in nearby domiciliary environments; thus, these areas must be monitored.

In many animal species, sex determination is accomplished by heterogamety i.e., one of the sexes produces two types of gametes, which upon fertilization will direct the development toward males or females. Both male ("XY") and female ("ZW") heterogamety are known to occur and can be easily distinguished when the sex-chromosomes are morphologically different. However, this approach fails in cases of homomorphic sex chromosomes, such as the sand fly Lutzomyia longipalpis s.l. (Psychodidae, Diptera), which is the main vector of visceral leishmaniosis in Brazil. In order to identify the heterogametic sex in L. longipalpis s.l., we did a whole-genome sequencing of males and females separately and used the "Y chromosome Genome Scan" (YGS) method to find sex-specific sequences. Our results, which were confirmed by PCR, show that L. longipalpis s.l. has XY system. The YGS method can be especially useful in situations in which no morphological difference is observed in the sex-chromosomes or when fresh specimens are not readily available.

Floral scent is a key mediator in plant-pollinator interactions. However, little is known to what extent intraspecific scent variation is shaped by phenotypic selection, with no information yet in deceptive plants. In this study, we collected inflorescence scent and fruit set of the deceptive moth fly-pollinated Arum maculatum L. (Araceae) from six populations north vs. five populations south of the Alps, accumulating to 233 samples in total, and tested for differences in scent, fruit set, and phenotypic selection on scent across this geographic barrier. We recorded 289 scent compounds, the highest number so far reported in a single plant species. Most of the compounds occurred both north and south of the Alps; however, plants of the different regions emitted different absolute and relative amounts of scent. Fruit set was higher north than south of the Alps, and some, but not all differences in scent could be explained by differential phenotypic selection in northern vs. southern populations. This study is the first to provide evidence that floral scents of a deceptive plant are under phenotypic selection and that phenotypic selection is involved in shaping geographic patterns of floral scent in such plants. The hyperdiverse scent of A. maculatum might result from the imitation of various brood substrates of its pollinators.

Skip oviposition is considered as an example of spatial bet hedging and involves a tradeoff between the benefit of reducing the risk of complete reproductive failure due to stochastic loss of a breeding site and the energetic and mortality costs associated with dispersal across several oviposition sites. Skip oviposition has been studied extensively for container-breeding mosquitoes but has never been studied with sand flies. By conducting a series of bioassays using solitary gravid females exposed to varying numbers of oviposition sites and to oviposition sites of variable quality at small (oviposition jar) and medium (free-flight cage) scales, we showed that sand flies exhibited skip oviposit at both scales. Specifically, with low-quality oviposition sites, females spread their eggs across several oviposition sites with total egg clutch size remaining constant and number of eggs per oviposition site decreasing with increasing number of oviposition sites. With variable quality sites, sand flies biased their oviposition to sites containing increasing levels of organic matter (OM), but also laid eggs in poor-quality sites; a behavior consistent with spatial bet hedging. We also demonstrated that the presence of OM stimulated larger egg clutch size and increased percent skip oviposition. Skip oviposition was less frequent at the free flight cage scale. But, at this scale, females were shown to be stimulated to lay more eggs when in the presence of other females than when alone.

Genus and species of phlebotomine sand flies have been recorded and described in caves in Brazil, but no study has provided the food source used by sand flies in these environments. Herein, we identified the blood source used by sand fly species in caves located at "Quadrilátero Ferrífero" (QF), Minas Gerais state. Specimens were manually collected near or on anurans inside ferruginous caves in Serra do Gandarela National Park and Serra do Rola Moça State Park. Males and females were placed in vials with 70% alcohol and stored at -10°C. Females engorged, after specific identification, had DNA extracted and followed for PCR amplification using specific primers. Sequencing was analyzed in the GenBank and Barcode of Life. A total of 198 specimens were collected (107 females and 91 males), all of them belonging to species Sciopemyia aff. microps (88.89%), Sciopemyia sordellii (10.61%), or Martinsmyia oliveirai (0.50%). When it comes to the females, 89 were S. aff. microps and 18 S. sordellii. Nineteen engorged females of S. aff. microps were analyzed and most of them (n=18) presented blood from Bokermannohyla martinsi and one contained blood from Scinax fuscovarius. The blood present in engorged females of S. sordellii (n=4) was from B. martinsi. Sciopemyia genus specimens are commonly found in collections carried out inside natural caves, but this was the first study to prove that females of this genus feed on cold-blooded animals in nature. HIGHLIGHTS: • Here we proved that sand flies feed in cold-blooded animals in in Brazilian caves. • Females of the Sciopemyia genus were for the first time found feeding in natural habitats. • Anurans of the family Hylidae were identified as source by molecular analyzes. • Insect bloodmeal identification can help assessing the fauna in several biomes. • This is the first record of S. aff. microps in caves of Brazil.

Cutaneous leishmaniasis is a crucial vector-borne disease caused by various species of Leishmania and is transmitted by several species of sandflies. The present study was conducted to describe sand fly fauna on vectors of leishmaniasis and performing molecular identification of Leishmania isolates from them on the Iran-Iraq border. Entomological surveys were done from May to October 2016-2018 in 2 counties (Mehran and Dehloran) of Ilam province, west of Iran. Sandflies were collected by 40 Sticky Traps at each station. Samples were mounted for species identification using morphological characters of the head and abdominal terminalia. DNA was extracted from Phlebotomus papatasi females, and Leishmania isolates were identified through PCR on minicircle kDNA, followed by sequencing. A total of 5592 sandflies including 2 genera of Phlebotomus and Sergentomyia comprising 8 species of sand flies were detected. Leishmania major infection was detected in 3.33% of 300 tested female sandflies. Phlebotomus papatasi was predominant in outdoor and indoor resting places. Phlebotomus papatasi was determined as dominant vector of Leishmania major infection in Mehran and Dehloran counties, West of Iran. It seems the composition of sandfly species in the study area is almost similar to the other parts of Iran. A detailed description of the epidemiology and ecology of Phlebotomine sand flies needs to be established to accomplish effective vector control programs.

In phlebotomine sand fly, the topic of the factors stimulating or inhibiting egg hatching has been largely ignored. In this study, we evaluated the hypothesis that pharate neonate sand flies are able to regulate their hatching time adaptively in response to cues indicative of the presence of food or potential competitors. In this study, we evaluated the independent and combined effects of organic matter (OM) extract and proximity to conspecific eggs on the hatching proportion of Phlebotomus papatasi eggs. In one set of larval rearing jars, we introduced 16 eggs into a small hole in the center of a plaster base of the jar. In another set, we introduced a single egg into each small hole of a 4-by-4 symmetrical array. To one set, we added an aqueous OM extract, and to the other, we added deionized water (DI). OM stimulated egg hatching while egg clustering slightly inhibited egg hatching. Results of this experiment are biologically important because they show, for the first time, that pharate sand fly neonates are able to adaptively regulate their hatching time in response to external cues.

Seoda cavernicola sp. nov. and Psychoda glamocensis sp. nov., are new species and cave dwellers from Bosnia and Hercegovina, and Croatia. Adults of S. cavernicola are pale and small; the eye bridge is reduced, ommatidia irregularly arranged, epandrium with a pair of setose excrescences. The eye bridge of P. glamocensis is likewise reduced with 2 or 3 irregularly ordered facet rows, palpus segments of some individuals are malformed; its closest relative is Psychoda alticola Vaillant based on the morphology of male and female genitalia as well as on COI barcodes.

Bruchomyiinae is one of seven subfamilies of Psychodidae. This small group comprises fewer than 60 extant species distributed mainly in tropical and sub-tropical regions (Wagner Stuckenberg 2016). All life stages of these flies are closely tied to forest environment (Fairchild 1952; Stuckenberg 1962) and as a result, bruchomyiines are frequently preserved in fossil resins. The first fossil member of this subfamily was described from Baltic amber (45 Ma) (Meunier 1905) and Burmese amber (100 Ma) is the oldest fossil resin in which representatives of Bruchomyiinae have been found (Stebner et al. 2015; Wagner 2017; Skibinska et al. 2019). Due to the relative scarcity of this subfamily in Baltic amber, any new specimens are of great interest. Up till now, there are only 12 bruchomyiine species described from Baltic amber. Wagner (2017) completed a revision of fossil Bruchomyiinae and proposed the genus Hoffeinsodes to include the species having male genitalia with gonocoxites fused with hypandrium. In this genus, he classified six species reported exclusively from Eocene Baltic amber: H. bifida Wagner, 2017, H. cubicula Wagner, 2017, H. longicauda Wagner, 2017, H. obtusa Wagner, 2017, H. reducta Wagner, 2017 and H. hoffeinsi (Wagner, 2006) transferred from Nemopalpus (Wagner, 2006). Here, we describe a new species belonging to this genus.

RESUMO

We documented and analyzed moth fly occurrence and spread of multidrug-resistant bacteria in a tertiary care hospital in Germany. The moth flies (Clogmia albipunctata) bred in the sewage system, then moved into the hospital, carrying biofilm and multidrug-resistant bacteria on their feet. Subsequently, the hospital developed a pest control protocol.

Tonnoiriella Vaillant, 1971 was one of the genera not treated by F. Vaillant for his revision in Die Fliegen der paläartktischen Region. Here we provide a genus diagnosis, re-describe known species based mainly on type material, and provide descriptions and figures of the West-Palearctic species. New species described are T. aurasica sp. nov., T. italiae sp. nov., T. ikariae sp. nov., T. andradei sp. nov., T. rhodesica sp. nov., and T. goncalvesi sp. nov.; T. androsica Vaillant, 1978, T. orientalis Vaillant, 1978, and T. pallidipenis Vaillant, 1978 are nomina nuda; finally 21 species are known to date from Europe and North Africa. The ejaculatory apodeme (basiphallus) is asymmetric and bilobed, lobes are stacked; one or both lobes are connected with asymmetric, cross positioned (distiphallus) sclerites. Operating of the open-close mechanism of the aedeagus is explained. Information on larval ecology is provided and the systematic position of the genus is discussed.
